Ogólnie - nie warto, nie będzie aż tak widocznych zmian.
Poza tym przy GABA jest problem z barierą krew-mózg, a melatonina obniża poziom teścia.
Nie wspominam już o problemach z legalizacją, które mogłyby zaistnieć.

Kurcze Grasik serio melatonina obniża testrosteron? No ostatnio widziałem badania, że ma wpływ na zwiększanie hormonu wzrostu i już myślałem, że zapodam na przyszły cykl masowy ale widze lipa?

Powiem w ten sposób - spotkałam się z badaniami to potwierdzającymi. Jednak jak ze wszystkim - znajdą się i przeciwnicy tej tezy. Nie wiem, czy wyszło coś nowego w tej kwestii, nie sprawdzałam.
Swoją drogą najlepszą rzeczą, którą brałam "na spanie" był... Venom Lean Dreams (też zawiera melatoninę i kilka ziółek). Spało się aż za dobrze (zaspałam do pracy ;)).

Abstract

OBJECTIVES: We have investigated the changes in serum luteinizing hormone (LH), follicle stimulating hormone (FSH) and testosterone levels together with testicular histology in both pinealectomized (PNX) and intact rats.

MATERIAL and METHODS: Twenty-one animals were PNX and allowed to recover for two months. Group I was assigned as PNX, group II PNX+melatonin and group III PNX+Human Chorionic Gonadotropin (HCG). Rats in group IV were sham PNX (S-PNX). An intact group of animals was s.c. injected with melatonin (0.5 mg/kg/day), another group with a combination of melatonin+HCG (5000 IU/kg/day) for seven days. Controls received saline alone (1 ml/kg). At the end, all animals were decapitated and blood samples obtained. Serum LH and FSH levels were determined by Radioimmunoassay, testosterone values by Chemiluminescent Enzyme Immunassay. Testicular tissue was collected and processed for light microscopy.

RESULTS: Serum LH levels were increased following PNX, but no such increases were seen in testosterone. In the PNX+melatonin group, serum LH and testosterone values were found to be similar to those of S-PNX group. HCG supplementation to PNX rats resulted in significant decreases in LH (p<0.005), but increased testosterone levels (p<0.001). Melatonin administration to intact animals significantly decreased both LH and testosterone levels (p<0.01). Co-administration of HCG+melatonin resulted in significant decreases in LH (p<0.001) and increases in testosterone levels (p<0.01). Serum FSH values did not show significant changes among groups. Only HCG administration significantly reduced FSH levels (p<0.01).

CONCLUSIONS: Our results suggest that melatonin inhibits testosterone secretion by acting at hypothalamo-pituitary axis. There is a functional relationship and feedback regulation between the pineal gland and the testes.


A novel mechanism for the melatonin inhibition of testosterone secretion by rat Leydig cells: reduction of GnRH-induced increase in cytosolic Ca2+

The site of inhibition, by melatonin, of GnRH-dependent testosterone secretion was investigated in adult rat Leydig cells cultured in vitro. The various effects downstream of the binding of GnRH to its own receptor were isolated and mimicked by specific drugs. Testosterone secretion was then evaluated after 3 h stimulation with GnRH, thapsigargin (1 microM), phorbol-12-myristate-13-acetate (100 nM), arachidonic acid (20 microM), and ionomycin (1 microM) in the presence or absence of melatonin (215 nM). The effect of melatonin on the GnRH-induced changes in cytoplasmic calcium concentration ([Ca(2+)](i)) was also studied, using Fura-2 as fluorescent Ca(2+) indicator. Melatonin attenuated the increase in [Ca(2+)](i) and inhibited the testosterone secretion induced by GnRH, but not that induced by ionomycin. Both ionomycin and thapsigargin potentiated GnRH-induced testosterone secretion; however, ionomycin, but not thapsigargin, partially prevented the inhibitory effect of melatonin on cells stimulated with GnRH. The effect of melatonin was probably dependent on the binding of melatonin to its Gi-protein-coupled receptor, as the inhibitory effect on GnRH-induced secretion was supressed in cells pretreated with pertussis toxin in a concentration of 180 ng/ml for 20 h. Assay of 17-hydroxy-progesterone showed that, irrespective of the treatment, cells cultured with melatonin secreted greater amounts than controls. We conclude that melatonin reduces GnRH-induced testosterone secretion by 1) decreasing [Ca(2+)](i), through impairment of the GnRH-dependent release of Ca(2+) from intracellular stores and 2) blocking 17-20 desmolase enzymatic activity, an effect that occurs irrespective of changes in [Ca(2+)](i).

The androgen milieu plays an important role in the control of immunity, and melatonin (MLT) is known to modulate the immune response as well. Our recent studies have focused on the role of MLT in controlling androgen secretion from rat testis. The presence of MLT receptors, their modulation, and the effect of MLT on steroidogenesis have been studied on Percoll-purified rat Leydig cells (LCs) cultured in vitro. MLT receptors present in adult rat LCs (Bmax = 46.7 &#177; 3.5 fmol/mg protein; Kd = 88.7 &#177; 6.2 pmol/l) are coupled through a pertussis toxin-sensitive G-protein and are downregulated by prolonged exposure to MLT itself. When acutely added for three hours, MLT (0.4-400 nM) inhibits T secretion in response to LH, forskolin, and GnRH, but sensitizes cAMP-dependent T secretion when present in the preincubation media (16 h). The acute inhibition of adenylyl cyclase and the block of 17-20-desmolase (with increased 17-OH-progesterone) and of calcium release from the intracellular stores, followed by sensitization of the adenylyl cyclase activity in the long term, are described as mechanisms of MLT action. Ultimately, our studies suggest that MLT is likely to control the immune response not only through its direct effects, but also by finely modulating the androgen milieu.
